SUBSTANCE: invention relates to the field of biotechnology. One-time PCR chip is proposed. Chip includes made of polycarbonate or cyclic polyolefins and having a body plate. Six reaction cells are located in the housing. Each reaction cell contains a reaction chamber, an expansion chamber connected to the reaction chamber, a filling hole and an outlet hole on the front surface of the chip, a filling and outlet channels and a sealing valve. Sealing valve includes a pusher and an elastic sealing element.

EFFECT: invention provides real-time PCR of up to 10 copies of the plasmid per reaction, complete elimination of contamination by reaction products when required conditions for heat transfer are met, fluorescence is passed, inertness to components of the reaction mixture.
