FIELD: measuring; biotechnology.
SUBSTANCE: invention relates to measuring equipment, namely to a device for identifying the causative agent of tuberculosis Mycobacterium tuberculosis and non-tuberculosis mycobacteria for use in medicine, molecular biology and biotechnology for the purpose of differential diagnosis of tuberculosis and determination of resistance of mycobacteria to antibacterial preparations. Device for automatic isolation, purification and amplification of DNA sections of Mycobacterium tuberculosis with registration of results in real time consists of a disposable cartridge, an ultrasonic generator, a UV radiation source, analyser, lodgement, cover, drive shaft, permanent magnet, rotary valve, which contains a cylinder with a movable piston and engages with the movable rod of the analyser when the cartridge is installed, which is a device capable of interacting with the disposable cartridge and controlling the processes of DNA extraction and subsequent PCR with detection of the fluorescent signal in real time inside the closed volume of the cartridge. Analyser includes at least one UV radiation source located inside the analyser, which provides disinfection of internal cavities of the analyser. Ultrasonic radiation source adjoins the wall of the container in the cartridge, which is made in the form of a convex lens.
EFFECT: creation of a device which is able to detect presence of Mycobacterium tuberculosis in biological samples, determine the resistance of the tuberculosis agent to antimicrobial preparations by simultaneous amplification of several specific DNA regions in real time in automatic mode for not more than three hours.
