SUBSTANCE: invention relates to the field of biotechnology, in particular to a method for modulating the expression of two or more target genes in a eukaryotic cell, involving the use of orthogonal RNA-directed non-nuclease-active DNA-binding proteins of Type II CRISPR system, which bind to two or more target genes and are directed by two or more guide RNAs. Methods for modifying one or more target genes in a eukaryotic cell are also disclosed, also involving the use of orthogonal RNA-directed non-nuclease-active DNA-binding proteins of Type II CRISPR system. The invention also relates to a eukaryotic cell expressing genes that are modulated by the above methods.
