SUBSTANCE: invention relates to the field of biotechnology, in particular to a eukaryotic cell containing a first foreign nucleic acid encoding two or more guide RNAs that are complementary to two or more corresponding target genes, a second foreign nucleic acid encoding two or more orthogonal RNA-guided nucleases or nickases of the CRISPR type II system, which, respectively, bind to two or more target genes and are guided by two or more guide RNAs, where each guide RNA and orthogonal RNA-guided nuclease or nickase of the CRISPR type II system are members of a co-localization complex on the corresponding target gene.
EFFECT: invention is effective for splitting and introducing a single-stranded break into target genes.
