MICROFLUIDIC METHODS AND CARTRIDGES FOR CELL SEPARATION Russian patent published in 2020 - IPC B01L3/00 C12N5/02

Abstract RU 2732235 C2

FIELD: biotechnology.

SUBSTANCE: invention relates to identification and preferential selection of cells on the surface of which a protein of interest is exposed, a cell selection cartridge based on the exposure level, an integrated system for cell selection, based on the level of exposure, a cartridge and an integral system for selecting cells based on the level of exposure and optionally secrete proteins from the population of cells, containing said cells exposing and optionally secreting said protein, as well as a kit for identifying and preferentially selecting cells on the surface of which a protein of interest is exposed. Method comprises providing a sample containing said cells; providing granules for gripping, containing one or more affine groups, and optionally carrier granules, wherein said affinity group(s) are adapted for cell binding, on the surface of which said protein is exposed, where grips for gripping are superparamagnetic granules, and carrier granules are ferromagnetic granules. Said cells are then mixed with said granules for capture and optionally with said carrier granules, wherein said affinity group of grips for binding binds said protein, which is exposed on surface of cells, to obtain magnetic-labeled cells (MMK) containing a magnetic mark. Method then includes separating, for example, during at least one washing step, cells not marked with magnetic particles from said MMK, and identifying and preferentially selecting cells, on the surface of which said protein is exposed.

EFFECT: invention widens the range of equipment.
