SUBSTANCE: group of inventions relates to biotechnology. Composition for increasing specific productivity of cell line-producers of antibodies based on CHO cells contains the following initial components per 1 l: L-glutamine in amount of 1.168 g, Pluronic F-68 in amount of 1 g, insulin in amount of 0.01 g, transferrin in amount of 0.0055 g, sodium selenite in amount of 0.0000067 g, hydrolyzate of peas proteins in amount of 0.06 g and IMDM medium - balance up to 1 l. Method of preparing the composition involves the following steps. Proteins are prepared by mixing initial components in the form of their solutions in buffer with initial concentration of 0.7 mg/ml. Proteins are extracted and centrifuged. Perform qualitative and quantitative analysis and subjected to hydrolysis with papain. Reaction is stopped with subsequent quantitative analysis. Precipitate is removed, and the extracted protein solution is filtered on the membrane to cut off hydrolyzate components with molecular weight higher than 5 kDa.

EFFECT: group of inventions provides higher specific productivity in cultivation of CHO cells in serum-free serums based on IMDM.
