Invention discloses a method, where the nucleic acid amplification reagent contains a set of primers, which amplify a target sequence and a complementary sequence, and a signal-generating substance which generates or quenches the signal in response to amplification, and the signal-generating substance generates a signal in a state in which it is associated depending on the sequence, and quenches the signal in a state in which it is not associated, or quenches the signal in a state, in which it is connected depending on sequence, and generates signal in state, in which it is not connected, and generation and suppression of signal are reversible.
