SUBSTANCE: group of inventions relates to the field of biotechnology. A sensor for detecting single molecules, a labeled nucleotide, a kit, a sensor system and a method for sequencing nucleic acids are proposed. The sensor includes two electrodes and a modulated electrically conductive channel connected to the electrodes. The modulated electrically conductive channel comprises a polymer in the form of a partially double-stranded nucleic acid. The polymer includes a gap in the first of the polynucleotide chains, which lacks nucleotide bases, and a plurality of nucleotide bases exposed in the gap region of the second of the polynucleotide chains. The labeled nucleotide contains a nucleotide, a binding molecule, and a switch strand, where the switch strand includes a strand of nucleotides including complementary bases exposed in the gap region of the sensor. The kit contains a substrate, two electrodes and a polymer solution. The sensor system contains a flow cell and a sensor. The method includes introducing a polynucleotide chain - a template into a sensor having a polymerase, introducing labeled nucleotides into the sensor and detecting a sensor response.

EFFECT: inventions provide sensitivity of the sensor at the level of a single molecule.
