FIELD: molecular biology.
SUBSTANCE: present invention relates to molecular biology, in particular to a method for loading exosomes with small non-coding RNAs, for the possibility of their subsequent targeted delivery to individual tissues of living organisms. To implement the method, exosomes obtained from urine or human peripheral venous blood and small non-coding RNA are mixed in phosphate-buffered saline (PBS) in a ratio of 1*10∧10÷5*10∧10 exosomes and 10*10∧12÷50*10∧12 copies of small non-coding RNA per 1 ml of solution. Then the resulting mixture is kept in a thermostat at 35-40°С for 14-18 hours.
EFFECT: present invention ensures maximum loading of exosomes obtained from human biological fluids with small non-coding microRNAs, which will further allow the loaded exosomes to be used as transport structures for the delivery of therapeutic agents into tumor cells of living organisms.
