FIELD: medicine.
SUBSTANCE: invention relates to medicine, namely to laboratory diagnostics, and can be used in the implementation of sample preparation for the identification of ethyl glucuronide in the blood. A biosubstrate sample is prepared and its chromatography-spectrometric study is carried out with the registration of the mass spectrometer signal in the form of a profile of the peaks of the analytes on the chromatogram, followed by the determination of the belonging of each peak to the analyte and comparison with the reference analytical characteristics of the desired substance. To prepare a biosubstrate sample, 200 mcl of blood is injected into a flat-bottomed vial and dried at 100°C for 7 minutes in order to denature proteins and remove moisture from the sample. Then it is cooled to room temperature, 250 mcl of methanol is added, covered with a lid, and shaken on a vibrating mixer for 2 minutes. The resulting extract is transferred to a vial and 100 mcl of deionized water is added. Then 5 mcl of the extract is injected into the chromatograph.
EFFECT: method makes it possible to increase the accuracy of identification of ethyl glucuronide in cadaveric blood or blood sampled from a living person by conducting a chromatography-spectrometric study with registration of a mass spectrometer signal in the form of a peak profile of analytes on a chromatogram, in which the most important operation, which significantly determines an increase in the identification accuracy , is the drying of the sample at 100°C for 7 minutes, since in this case the protein mud coagulates and its effect on the identification accuracy is sharply reduced.
