FIELD: biology.
SUBSTANCE: group of inventions relates to a recombinant bacterium constitutively producing monophosphoryl lipid A (MLA) not conjugated to the 2-keto-3-deoxy-D-manno-octulosonate (Kdo) grouping, and to a method of production of MLA not conjugated to the Kdo grouping using the described bacterium. Proposed is a recombinant bacterium constitutively producing MLA not conjugated to the Kdo grouping, wherein said recombinant bacterium has increased expression of the gene encoding the lipid A 1-phosphatase polypeptide (LpxE) as compared to the parent bacterial cell. The chromosome of the bacterium also contains deletion, reading frame shift mutation or substitution of the polynucleotide encoding undecaprenyl pyrophosphate phosphatase (Und-PP phosphatase), deletion, reading frame shift mutation or substitution of the polynucleotide encoding phosphatidylglycerophosphate phosphatase (PGP) and deletion, reading frame shift mutation or substitution of the endogenous polynucleotide encoding 3-deoxy-D-manno-octulosonic acid (KdtA) transferase. The method of producing MLA not conjugated to the Kdo grouping includes culturing said bacterium producing a culture, collecting the bacterium from the culture, and isolating said MLA from the collected bacteria.
EFFECT: group of inventions provides efficient production of said MLA using a bacterium constitutively producing MLA.
