FIELD: medicine; laboratory diagnostics.
SUBSTANCE: invention relates to medicine, namely to laboratory diagnostics, it can be used for multicolored immune-cytochemical diagnostics of paraneoplasia of the cervix uteri. A cell suspension of the clinical material is obtained, stain with a fluorescent dye is performed, followed by fluorescence microscopy. The cell suspension is added to a mixture of antibodies specifically directed to the proteins P16/P16INK4a, Ki67 and EpCAM/clone BerEp4, labeled with fluorescent dyes of different wavelengths. The mixture includes antibodies to the Ki67 protein conjugated to fluorophore with a red-spectrum wavelength, antibodies to the EpCAM/clone BerEp4 protein conjugated to fluorophore with a green-spectrum wavelength, and antibodies to the P16/P16INK4a protein conjugated to fluorophore with a blue-spectrum wavelength. The incubation is carried out in a dark place at room temperature for 10-15 minutes. After incubation, fluorescence microscopy of the antibody mixture and the cell suspension is performed without waiting for drying. If at least one cell with triple immune staining for P16/P16INK4a, Ki67, and EpCAM/clone BerEp4 proteins is detected in the cytological material, paraneoplasia of the cervix uteri is diagnosed.
EFFECT: method provides for possibility of rapid, effective diagnostics of severe dysplasia and tumors of the cervix uteri due to possibility of rapid parallel determination of the co-expression of Ki67, P16 proteins by immune-cytochemistry and additional confirmation of the epithelial nature of cells by determining the expression of the EpCAM/clone BerEp4 protein on the surface membrane of the studied cells.
