FIELD: medicine and applied microbiology.
SUBSTANCE: invention relates to medicine and applied microbiology, namely to a method for determining the permeability of viral particles of a vaccine strain of poliomyelitis virus Sabin 1 of tissue and nonwovens. A method for determining the permeability of the vaccine strain of the poliomyelitis virus Sabin 1 of tissue and nonwoven materials by viral particles, in which initially a visual inspection and rejection of the test samples with mechanical damage affecting the permeability are carried out, the selected samples of the test samples are cut into flaps in the form of a circle with a diameter of 47 mm based on 3 flaps per study from each tissue sample, which are sterilized in glass Petri dishes in a steam sterilizer for 20 minutes at a temperature of 120±1.0°C and a pressure of 1 Pa, the obtained sterile flaps are sequentially placed in a pressure filtration device with a liquid container and a frit diameter of 47 mm as a filter element through which 10 ml of a suspension of the Sabin 1 polio virus vaccine strain diluted in saline, at a concentration of 105 viral particles/ml, is passed under a pressure of 1-6 atmospheres, the resulting eluate is processed using chloroform or sterilizing filter tips, then the processed eluate is examined for the amount of filtered viral particles, the permeability of tissue and nonwoven materials by viral particles of the poliomyelitis virus as the product of the ratio of the arithmetic mean number of viral particles found in eluates from three flaps of a selected sample to the number of viral particles found in the diluted viral suspension used in the study per 100 is calculated.
EFFECT: invention allows estimating with high reliability the permeability of tissues and nonwovens by viral particles of the vaccine strain of poliomyelitis virus Sabin 1.
