SUBSTANCE: invention relates to biotechnology and specifically to production of the fauna vaccines, and can be used to determine concentration of 146S foot-and-mouth disease virus particles in non-inactivated raw materials for vaccine production. That is ensured by evaluating the number of viral RNA molecules recovered after virion immune capture. RNA is isolated from immune complexes "146S particle-antibody". Degree of purity of RNA extracts is assessed by spectral analysis and gel electrophoresis. Series of dilutions of a positive standard of foot-and-mouth disease RNA virus with a number of molecules corresponding to concentrations of 146S particles from 0.1 to 4.0 mcg/ml with a dilution interval of 0.1 mcg/ml is prepared. Number of RNA molecules is calculated from the absorption spectrum. Dependence of concentration of 146S particles and the number of RNA molecules is obtained in the form of a positive linear model. On the basis of the received linear model values of concentration 146S of foot-and-mouth disease virus particles in samples of raw materials are calculated.
EFFECT: invention enables fast and high-reliability determination of 146S concentration of foot-and-mouth disease virus particles in non-inactivated raw material for vaccine production.
