FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, in particular to a method for molecular genetic identification of unknown wood samples in coniferous plant species, when using DNA samples isolated directly from log wood in the place of its harvesting or after transportation in PCR. The method includes 6 stages: collection of unknown wood samples; isolation of DNA samples from them; ISSR (Inter Simple Sequence Repeats) analysis of DNA polymorphism using PCR; selection of the most genetically heterogeneous DNA samples tested; determination (sequencing) of nucleotide sequences from one to three polymorphic loci in each tested species of coniferous plants in the research region; identification of DNA fragments characteristic of the genus (generic), for the species (specific) and combinations of polymorphic population ISSR-PCR, and in some populations SNP markers as well; comparison (identification) of the identified DNA fragments with previously established identification DNA fragments for natural populations of the tested species in the research region. The invention makes it possible to effectively identify unknown wood samples from coniferous plant species and establish the correspondence or absence of DNA fragments of felled trees to identification DNA fragments of trees growing in the natural population of the species in the research region. The invention makes it possible to establish the identity of objects with high accuracy, provides for comparison (identity) of test samples, that is, a network of DNA fragments amplified with DNA samples from wood, with already existing studied objects, in this case with generic, species and combinations of population identification DNA fragments identified during the preliminary molecular genetic analysis of natural populations of coniferous plants in the research region.
EFFECT: extension of the range of methods for softwood research.
