SUBSTANCE: invention relates to medicine and concerns a method for determining steroid hormones in human urine, including the preparation of the analyzed sample by enzymatic hydrolysis, extraction, evaporation of the extract, dissolution of the dry residue and subsequent chromatographic separation, using reverse-phase high-performance liquid chromatography with quadrupole-time-of-flight mass spectrometric detection, while the sample preparation is carried out by introducing urine containing the internal standard methyltestosterone into the sample, a phosphate buffer solution with the β-glucuronidase E. Coli enzyme, followed by incubation of the resulting mixture, and a mixture consisting of chloroform and a dispersant is introduced, mixed, centrifuged, after phase separation, the lower organic layer is selected, evaporated in nitrogen current and a diluent is added to the dry residue, while acetone in the amount of 500 µl is used as a dispersant, and an aqueous solution of hydroxylamine and methanol 1:1 is used as a diluent, the resulting mixture is incubated for at least 90 minutes at a temperature of at least 70°C.

EFFECT: invention provides an increase in the sensitivity of the determination of steroids.
