TEST SYSTEM FOR DETECTING SPECIFIC NUCLEOTIDE SEQUENCES CHARACTERISTIC OF THE DETECTED MICROORGANISMS OR VIRUSES, METHOD FOR APPLICATION OF THE TEST SYSTEM (OPTIONS) Russian patent published in 2022 - IPC C12N15/86 C12Q1/70 C12Q1/6806

Abstract RU 2769572 C1

FIELD: molecular biology.

SUBSTANCE: invention relates to the field of molecular biology, virology and biotechnology. A test system is described for detecting specific nucleotide sequences characteristic of the detected microorganisms or viruses by amplification of nucleic acids of microbes or viruses, containing DNA polymerase with chain-displacing activity, primers for loop isothermal amplification, deoxyribonucleotides, salts, as well as detergent Triton™ X-100, bovine serum albumin, dithiothreitol, ethylenediaminetetraacetic acid, while the concentration of substances in the final liquid for the nucleic acid amplification reaction is: detergent Triton™ X-100, from 10 to 150 g/l, bovine serum albumin from 1 to 20 g/l, dithiothreitol from 6.25 to 18.75 mmol/l, ethylenediaminetetraacetic acid from 0.5 to 1.5 mmol/l. Also described is a test system for detecting specific nucleotide sequences characteristic of the detected microorganisms or viruses, by amplification of nucleic acids of microbes or viruses, by amplification of nucleic acids of microbes or viruses, containing DNA polymerase with chain-displacing activity, reverse transcriptase, primers for loop isothermal amplification, deoxyribonucleotides, salts, and detergent Triton™ X-100, bovine serum albumin, dithiothreitol, ethylenediaminetetraacetic acid, at the same time, the concentration of substances in the final liquid for the nucleic acid amplification reaction is: detergent Triton™ X-100 from 10 to 150 g/l, bovine serum albumin from 1 to 20 g/l, dithiothreitol from 6.25 to 18.75 mmol/l, ethylenediaminetetraacetic acid from 0.5 to 1.5 mmol/l. In addition, a method is presented for using these test systems to identify specific nucleotide sequences characteristic of the detected microorganisms or viruses by amplification of nucleic acids of microbes or viruses, where amplification of viral or microbial nucleic acids is performed by loop isothermal amplification at a temperature from 55 to 69°C.

EFFECT: invention can be used for amplification of nucleic acids (NC) of various origins.
