FIELD: medicine.
SUBSTANCE: invention relates to medicine and concerns a method for diagnosing colorectal cancer (hereinafter – CRC) by quantitative assessment of the concentration of extracellular nanovesicles (hereinafter – ENV) secreted by colon epithelial cells and/or colon adenocarcinoma cells (hereinafter – cENV) as part of a total population of ENV. The method consists in isolating the total population of plasma ENV, assessing the size and concentration of ENV using trajectory analysis, forming immuno-sorbing complexes consisting of superparamagnetic particles (hereinafter – SPMP) with streptavidin and 5 antibodies to markers of intestinal epithelium differentiation CLRN3, GPA33, GCNT3, PIG-Y, Reg IV. SPMP-5AB complexes are collected on a magnetic tripod, supernatant with unbound antibodies is selected, and complexes are washed twice. Washed SPMP-5AB complexes are incubated with a sample of a total fraction of ENV. Formed SPMP-AB-cENV complexes are collected on the magnetic tripod, supernatant with unbound cENV is selected, and complexes are washed. cENV as part of SPMP-AB-cENV complex is labeled with antibodies to common exosomal markers CD63 or CD9 bearing a fluorescent label, cENV is isolated and quantified as part of immuno-sorbing AB-cENV-flAB complexes by flow cytometry, while in patients with CRC, the fraction of cENV is more than 5% of the total population of plasma ENV.
EFFECT: invention provides the diagnosis of CRC with sensitivity of 0.72 and specificity of 0.76.
