FIELD: biotechnology.
SUBSTANCE: invention refers to biotechnology, namely to extraction of exosomes from blood plasma. Method involves using SMP/AptCD63 complexes containing supermagnetic particles (SMP) with diameter of 1 mcm, coated with streptavidin and DNA aptamers AptCD63, modified with biotin molecule. Complexes are formed by incubation of 1 mcg of SMP coated with streptavidin, and 5 nanomoles AptCD63 modified with biotin molecule at room temperature for 30 minutes. Further, the SMP/AptCD63 complexes are washed from the unbound molecules of the aptamer with a phosphate-salt buffer, added to blood plasma with volume of 10-100 mcl and incubated for 10–12 hours at a temperature of +4 °C to form complex SMP/AptCD63/exosome.
EFFECT: invention enables performing exosomal blood plasma isolation for subsequent analysis of surface exosomatic proteins by flow cytometry.
