FIELD: microbiology.
SUBSTANCE: invention relates to a recombinant strain of Escherichia coli bacteria, a producer of methylcytosine-specific DNA glycosylase ROS1. A strain of Escherichia coli Rosetta 2(DE3)pET24b(+)-ROS1 bacteria is proposed, which is a producer of recombinant DNA glycosylase ROS1. This strain is obtained by transformation of Escherichia coli cells of the Rosetta 2(DE3) strain by the pET24b(+)-ROS1 plasmid having the nucleotide sequence SEQ ID NO:1, constructed on the basis of the pET24b(+) vector containing the f1 replication origin, the T7 phage promoter, the RBS ribosome planting site, NdeI and XhoI restriction sites, according to which a fragment of the gene is embedded, encoding DNA glycosylase ROS1 from Nicotiana tabacum tobacco, the His-tag histidine label, the T7 phage terminator, the kanamycin resistance gene Kan (R), as well as the lactose repressor lacI and the lactose operator.
EFFECT: invention provides for the production of recombinant protein ROS1 with a purity of at least 90%.
