FIELD: biotechnology.
SUBSTANCE: invention constitutes a method for producing an optimised nutrient medium for rootstock material (L-2) in vitro, wherein first test-tube plants are grafted and the single-node grafts are planted in an agarised nutrient medium containing macro- and microelements and vitamins based on the Murashige and Skoog nutrient medium. The resulting shoots are transplanted onto the Murashige Skoog nutrient medium with a content of 6-BAP of 1.7 mg/l, whereto a Ca(NO3)2*4H2O crystal hydrate 220 mg/l is added as a growth stimulant, at the following ratio of components, mg/l: ammonium nitrate 825 mg/l, potassium nitrate 950 mg/l, magnesium sulphate 185 mg/l, potassium hydrphosphate 85 mg/l, boric acid 6.2 mg/l, manganese sulphate 22.3 mg/l, cobalt chloride 0.025 mg/l, copper sulphate 0.025 mg/l, zinc sulphate 8.6 mg/l, sodium molybdate 0.25 mg/l, solution of chelated iron (iron sulphate 5.6 mg/l, disodium salt of ethylenediaminetetraacetic acid 37.3 mg/l), potassium iodide 0.83 mg/l, sucrose 15 mg/l, agar 8 mg/l; transplantation is performed after 3 weeks; in propagation, special attention is paid to the vitreousness or excessive water content of tissues, wherein the optimal conditions for the cultivation of L-2 are: temperature of 22 to 26 degrees, illumination of 2,500 to 5,000 lux, with a 16 hour photoperiod.
EFFECT: possibility of increasing the yield of plants up to 70%.
