FIELD: medicine.
SUBSTANCE: present disclosure relates to medicine, in particular to a method for preparation of RNA-lipoplex particles for RNA delivery to target tissues after parenteral administration and a composition containing such particles. For implementation of the specified method, a solution containing RNA and having ionic strength corresponding to sodium chloride at a concentration from about 50 mM to about 300 mM is mixed with a solution containing cationic liposomes under controlled mixing conditions. At the same time, the combined flow of the first solution and the second solution is characterized by Reynolds number of more than 300. As a result, RNA-lipoplex particles are obtained, having an average diameter, which is in the range from about 300 nm to about 500 nm, and having a polydispersity index of less than 0.2.
EFFECT: present invention allows for an increase in the efficiency of translation of RNA into peptide or protein, which it encodes, when delivering RNA to a target cell, as well as an increase in the stability of compositions during storage without significant loss of product quality, in particular, biological activity of RNA.
