FIELD: biotechnology.

SUBSTANCE: invention is a method for in vitro preservation of vegetative material, where modifications of previously developed protocols are used. The method consists in aseptic cultivation of plants under optimal conditions, isolation of explants for storage, and their transfer to special nutritional media in storage vessels, preservation of vessels with samples of vegetative material under conditions of growth inhibition. At the same time, the method consists of sequentially performed stages of: in vitro cultivation of vegetative material under conditions optimal for growth, preservation of samples of vegetative material on modified nutritional media added with 3-9 mM of calcium gluconate – C12H22CаO14 or calcium nitrate, 6-10% sucrose, 4-6 g/l of mannitol, 0.2-0.5 mg/l of 6-benzylaminopurine, 0.01-0.05 mg/l of indolyl butyric acid, 0.2-1 mg/l of paclobutrazol of [1-(4-chlorophenyl)-4,4-dimethyl-2(1,2,4-triazole-1-yl)pentane-3-ol], and 10-12 g/l of agar-agar, at +1-20°C in the light (4-16 hour day with illumination of 0.5-1.0 klx) or in the dark in the presence or absence of silver thiosulfate in capsules of calcium alginate. Growth of vegetative material is in vitro restored on agarized nutritional media added with 3% of sugars (1% of glucose and 2% of sucrose), 0.5 mg/l of 6-benzylaminopurine, and restored samples are reproduced on specialized nutritional media for active growth.

EFFECT: claimed method allows for in vitro preservation of potato shoots for up to 8 years.
