SUBSTANCE: inventions relates to the field of measurements based on differential detection of light from a sample in a flow, to flow cytometry. Light detection systems c include a flow cell configured to transfer a sample to the flow cell, a light source configured to irradiate the sample in the flow cell, and a detection system having an optical adjustment component, and a detector configured to differentially detect light from flow cell without diffuser bar. The systems are configured to differentially detect light by modulating one or more components of the optical adjustment component or detector. Methods for differential detection of light from a sample in a flow stream by means of a detector unit are described. Disclosed are kits having two or more components for use in the claimed systems.
EFFECT: improved cell sorting accuracy, improved particle collection, reduced energy consumption, particle charging efficiency, more accurate particle charging, and improved particle deflection during cell sorting.
