FIELD: biotechnology industry.

SUBSTANCE: invention relates to biotechnology industry in particular, to collagen hydrate solution preparation method. The method includes taking freshwater scaleless fish skin, cleaning them from muscular tissue cuts, fins and mechanical impurities, primary washing by a solution containing 1 wt. % of food surfactants in water and food surfactants solution to raw material ratio of 2 l solution to 1 kg of raw material, respectively, under mechanical agitation in a drum-type system with the drum rotation speed of at least 400 rpm at room temperature. Dermal membranes are incubated in peroxide-alkaline solution containing 750 mmol sodium hydroxide and 300 mmol hydrogen peroxide at room temperature until skin discoloration at a solution: raw material ratio of 2.5 l solution : 1 kg raw material, respectively. Then freshwater hydrobionts dermal membranes are washed with running water at 8-12°C to remove acids and alkalis at a mass ratio of dermal membranes : water : 1 : 3, respectively, with constant stirring; the operation is repeated three times, holding at constant stirring for 10-15 minutes at each stage. After that the dermal membranes are incubated in 117 mmol weak organic acid solution at pH of 4 for 48 hours under the constant influence of 100 kHz ultrasound. Then the liquid phase is removed by means of draining, distilled water is added to the received semi-finished product in a range of weight ratios semi-finished product to water - 1:31:5, respectively, and homogenization is carried out using a paddle type homogenizer with 400 W capacity for 5 minutes at a knife rotation velocity of 600 rpm to obtain a dispersion. The resulting dispersion is filtered using a 0.45 µm pore diameter filter. A cold sterilisation with ultraviolet light is performed and the dispersion is packaged.

EFFECT: invention provides increased yield of native collagen molecules with retained fibrillar structure while reducing process time.
