METHOD FOR OBTAINING AUTOLOGOUS REGULATORY T-LYMPHOCYTES BY EX VIVO CULTIVATION IN PRESENCE OF HUMAN CHORIONIC GONADOTROPIN Russian patent published in 2023 - IPC C12N5/783 A61K35/14

Abstract RU 2791738 C1

FIELD: biotechnology; cellular biotechnology.

SUBSTANCE: objective of the invention is to develop a composition of culture medium factors intended to improve the technology of cultivating CD4+-lymphocytes, leading to their effective differentiation into T-regulatory cells (Treg), expressing the FOXP3 transcription factor, which indicates the suppressor function of the resulting Tregs. To solve this problem, human chorionic gonadotropin was added to the composition of the traditionally used medium for cultivating CD4+ cells, among the physiologically significant functions of which its ability to cause mild immunosuppression is known. As a result, a method was developed for culturing and differentiating CD4+ cells into T-regulatory lymphocytes, which makes it possible to obtain a greater number of functionally active cells in a shorter cultivation time.

EFFECT: development of approaches to the treatment of a number of autoimmune disorders, as well as to reduce the risk of developing host-versus-graft reaction (“РХПТ”), which occurs when tissues and organs are rejected in cases of allogeneic transplantation.
