FIELD: biotechnology.
SUBSTANCE: invention relates to methods for manufacturing biomedical cellular products and methods for assessing the safe use of a cellular component included in their composition, and is intended for rapid assessment of the tumorigenic potential of cells planned for use as part of a biomedical cellular product. With the help of the system of intravital shooting of cells in culture and the subsequent analysis of the tracks of cell movements, control quantitative indicators of the motor activity of normal diploid fibroblasts and cells of tumour origin are established. The nature of the movement of the tested cells is evaluated, namely, the graphs of the average square of the displacement of cells depending on time and the graphs of the average square of the displacement depending on time are compared for a computer simulation of a random walk of point objects, built on the basis of data on the average speed and standard deviation of the average speed of the tested cells. Based on this comparison, a decision is made to classify the tested cells as either non-tumourigenic diploid cells or tumorigenic cells. Thus, an in vitro assessment of the tumorigenic potential of cells is carried out which can significantly reduce the probability of including tumour-forming cells in long-term and expensive preclinical animal experiments.
EFFECT: rapid and efficient detection of potentially tumorigenic cells both in primary culture, with their extremely limited number, and in long-term cultured cells.
