FIELD: biotechnology.
SUBSTANCE: perfusion filtration method for continuous cultivation of cell cultures in a bioreactor is presented, which includes the following steps: providing a culture medium containing cells in the bioreactor; selection of cultural fluid from the bioreactor in automatic mode with customizable software sampling of fluid; directing the culture liquid with a pump from the bioreactor to the filter holder containing filter elements, where cells are retained and returned to the bioreactor; when cells are retained on the filter elements, part of the liquid that does not contain cells is diverted to an automated control system for membrane valves with software, representing four separate filtrate discharge lines with pressure sensors that register pressure changes. The rate of pressure change shows the degree of contamination of the filter elements, with the option of controlling the frequency of opening and closing the membrane valves and starting the self-cleaning of the filter elements in automatic mode. Then, an additional sampling of the filtrate is carried out featuring automated control of the discharge of the filtrate with the replacement of the nutrient medium in the bioreactor with a fresh one.
EFFECT: increased density of cell cultures and increased yield of the product.
