FIELD: biotechnology.

SUBSTANCE: plasmid genetic construct pET21_AB_CoV-2_1.3 intended to obtain nanoantibodies against SARS-CoV-2, and a recombinant protein AB_CoV-2_1.3, which has the properties of a single-domain nanoantibody against SARS-CoV-2 are described. The plasmid genetic construct pET21_AB__CoV-2_1.3 ensures the expression in the prokaryotic system of E. coli of the recombinant protein AB_CoV-21.3 with the properties of a single-domain nanoantibody against SARS-CoV-2, has a size of 5,925 bp. and the nucleotide sequence SEQ ID NO: 1 and includes, in accordance with the physical and genetic map shown in fig. 1, the following elements: a sequence encoding the leader peptide pelB, which ensures the export of the protein from the cell; the nucleotide sequence AB_CoV-2_1.3 encoding the variable fragment of the heavy chain of the anti-SARS-CoV-2 nanoantibody; 6xHis — a sequence for the purification of nanoantibodies against SARS-CoV-2. The recombinant protein AB_CoV-2_1.3 has the properties of a single-domain nanoantibody against SARS-CoV-2, has a molecular weight of 19.7 kDa and the amino acid sequence of SEQ ID NO: 2.

EFFECT: creation of a plasmid genetic construct with a high and stable expression yield in the periplasm of E. coli bacteria of a recombinant protein with the properties of a single-domain nanoantibody against SARS-CoV-2.
