SUBSTANCE: method of determining pathogenic strains of Acinetobacter baumannii bacteria with multidrug resistance is proposed, including screening for the enzymatic activity of a nanoagent — a gold nanoparticle conjugate with a size of 20 to 32± 4 nm with the enzyme Dpo1053 depolymerase encoded in the genome of the lytic phage Acinetobacter baumannii AP22 or Dpo1432 depolymerase encoded in the genome of the lytic phage Acinetobacter baumannii vB_AbaP_AS12 immobilized on the surface of the nanoparticle. 10 µl of the nanoagent at a concentration of 1 nM is added to 100 µl of the bacterial sample and to the control sample containing Escherihia coli with OD600=0.2; then UV spectroscopy is carried out in 96-well plates, the optical density is measured for 1–3 min; in case of detecting shifts in the surface plasmon resonance to a longer wavelength region when a nanoagent is added to the analyzed strain in the first minutes of incubation, it is considered that the analyzed strain is a pathogenic strain of Acinetobacter baumannii.
EFFECT: invention provides an extension of the arsenal of agents for real-time recognition of pathogenic bacteria.
