FIELD: molecular diagnostics.

SUBSTANCE: invention concerns the detection and identification of DNA of the phytopathogenic fungi Alternaria solani Sorauer in biological material using polymerase chain reaction (PCR). A method for identifying the phytopathogenic fungus Alternaria solani by PCR is proposed, based on the use of species-specific primer pairs for PCR AltS1-F: 5' - AAG GAC САА ССС ATA AAC СТТ ТТТ GCA ATG-3' / AltS1-R: 5' - GAC ТТТ AAG GCG AGT CTC CCG CAA G-3', or AltS2-F: 5' - CTC GTC CGG CCG GAC СТТ CCT TTT AGC-3' / AltS2-R: 5' - TCT CGT AAG GTG CCG AGC GAG TCA GA-3'. The method involves isolating DNA from samples of biological material, carrying out a polymerase chain reaction with isolated DNA and species-specific primers, electrophoresis of PCR products in an agarose gel, their visualization and identification by comparison in size with marker DNA fragments of a molecular weight marker and/or amplicons of a control sample. The presence of PCR products of 325 or 463 nucleotide pairs in size when using primers AltS1-F and AltS1-R or AltS2-F and AltS2-R, respectively, in the sample indicates the presence of Alternaria solani in the sample under study.

EFFECT: method allows for early diagnosis of Alternaria blight on nightshade plants in the field, as well as carrying out phytosanitary control of the presence of this phytopathogen.
