SUBSTANCE: invention is a method of determining corrected color values from image data obtained during a base sensing cycle using a nucleic acid sequencer comprising an image acquisition system, one or more processors, and a storage device, comprising the following: (a) obtaining an image of a substrate containing a plurality of base sensing sites; nucleic acids, the portions having colors representing types of nucleic bases; (b) measuring color values of a plurality of regions from said substrate image; (c) storing the color values in a processor buffer of one or more sequencer processors; (d) retrieving phasing-corrected color values of a plurality of regions where said phasing-corrected color values were stored in the sequencer memory during the immediately preceding base recognition cycle; and (e) determining the corrected color values from the color values in the processor buffer and the phasing corrected color values stored from the immediately preceding cycle.

EFFECT: invention makes it possible to determine corrected color values from image data obtained using a nucleic acid sequencer during a base recognition cycle.
