SUBSTANCE: group of inventions relates to biotechnology. System and a computer-assisted method of determining a nucleotide sequence of polynucleotides are disclosed. System comprises a light source for causing emission of fluorescent radiation, a detector for detecting fluorescent radiations from a fluorophore attached to the nucleotide at a first wavelength and a second wavelength, as well as a processor for executing instructions. Method includes generation of fluorescent radiations by a light source, detection by a detector of intensity of fluorescent radiations with a first wavelength and with a second wavelength from a fluorophore attached to a nucleotide detector, colour correction of fluorescent radiation intensities and identification of a nucleotide.

EFFECT: inventions provide identification of all four nucleotide bases using a single light source and two different optical channels.
