SUBSTANCE: present invention refers to biotechnology. Disclosed is a method for sequencing nucleic acid molecules using physical and virtual unique molecular indices (UMI), where each UMI is an oligonucleotide sequence which can be used to identify an individual molecule of a double-stranded DNA fragment, wherein the physical UMI is presented in an adapter, which is attached to a fragment of double-stranded DNA found in the sample, and the virtual UMI is a unique sub-sequence in the original DNA molecule.

EFFECT: present invention can be further used in methods of analysing biological samples.
