SUBSTANCE: molecular genetic diagnostics and medicine, used in determining the hereditary predisposition to the development of diseases associated with carriage of the polymorphic variant rs6702742 (A>G) of the SERPINE1 gene of mRNA-binding protein 1. A method has been proposed for genotyping the polymorphic locus rs6702742 (A>G) of the SERBP1 gene in humans by real-time PCR using allele-specific fluorescent probes, involving PCR using specially selected primers (F: 5'-CCATCCCATAAGACATTATCTTGTAA-3' and R: 5'-GGGGAACTGTCAAAGACGAA-3') and probes with fluorophores (A-allele-specific fluorescently labeled probe 5'-(FAM) TATGGATTA A CCTCTAGTTA( RTQ1)-3' and G-allele-specific fluorescently labeled probe 5'- (ROX) TATGGATTA G CCTCTAGTTA( BHQ2)-3′) in an amplifier with fluorescence detection.
EFFECT: expanding the scope of methods for genotyping polymorphic variants of the SERBP1 gene, increasing accuracy and reducing analysis time.
