FIELD: biotechnology.

SUBSTANCE: proposed method of increasing the flavonoid-forming ability of buckwheat tissue culture in vitro includes cultivating microcuttings with an axillary bud on Murashige and Skoog medium containing ammonium nitrate, potassium nitrate, potassium phosphate monosubstituted, magnesium sulfate heptahydrate, calcium chloride dihydrate, iron sulfate heptahydrate, boric acid, manganese sulfate tetrahydrate, cobalt chloride hexahydrate, copper sulfate pentahydrate, zinc sulfate hepthydrate, sodium molybdate dihydrate, potassium iodide, thiamine chloride, pyridoxine chloride, sucrose, agar and water, obtaining test tube plants, their cuttings and subcultivation on Murashige and Skoog medium. A distinctive feature of the method is that microcuttings are gradually cultivated on Murashige and Skoog medium with the addition of copper sulfate pentahydrate in a concentration of 161 or 184 mg/l, zinc sulfate hepthydrate in an amount of 808–1,313 mg/l for 30–33 days, and then at III–V subcultivation on media without any toxicants, a study of the rutin content in buckwheat regenerants (without roots) is carried out using the chromatospectrophotometric method, lines with the highest level of flavonoid-forming ability are selected.

EFFECT: proposed invention will improve the flavonoid-forming ability of buckwheat tissue culture in vitro through the use of high concentrations of zinc in the nutrient medium, and increase the genetic diversity of the source material of F. esculentum.
