FIELD: medicine and clinical pharmacology.
SUBSTANCE: following is proposed: a method of inhibiting ABCB1 protein in vitro, including culturing a human colon adenocarcinoma cell line in Dulbecco's modified Eagle's medium containing glucose 4,500 mg/l, L-glutamine 4 mM, 15% bovine serum, 100 U/ml penicillin and 100 μg/ml streptomycin; after reaching 70–90% confluency, the cells are removed from the flask by adding a solution of 0.25% trypsin and 0.2% EDTA, seeded on a 24-well plate, and cultured for 48 hours; upon reaching the monolayer, mother aqueous colloidal solutions of modified nanoparticles gold-D-Lactose-Mercaptopropanoylhydrazide, gold-D-Galactose-Mercaptopropanoylhydrazide with a diameter of 18–21 nm are added to the medium until their concentration in the medium is 550 μg/ml and 300 μg/ml, respectively, followed by incubation for 8 hours.
EFFECT: invention expands the arsenal of methods of inhibiting the transporter protein to predict drug-to-drug pharmacokinetic interactions.
