FIELD: biotechnology.
SUBSTANCE: method is carried out as follows: germination of seeds of red clover samples differing in acid resistance is performed in Petri dishes on filter paper moistened with Gamborg B5 nutrient medium with 2.0 mg/l BAP (6-benzylaminopurine) with and without the addition of various concentrations of Al3+ and cultivation for 30 days until the formation of seedlings with epicotyls and roots without obtaining any morphogenic tissue. DNA is isolated by the SDS extraction method from a total sample of 30 seedlings with epicotyls and roots of each sample (bulk sample). Acid resistance is assessed based on a comparative analysis of differences in the nature of DNA amplification and acid resistance of F1 plant populations of each sample.
EFFECT: invention makes it possible to evaluate and select in vitro samples of red clover with increased resistance to environmental acidity using molecular genetic technologies.
