VOLTAMMETRIC METHOD FOR QUANTITATIVE DETERMINATION OF PREDNISONE IN BLOOD SERUM Russian patent published in 2024 - IPC G01N27/26

Abstract RU 2815787 C1

FIELD: chemistry.

SUBSTANCE: invention relates to analytical chemistry. Disclosed is a voltammetric method for the quantitative determination of the hormone prednisone in blood serum, involving the use of a three-electrode electrochemical cell containing a Britton-Robinson buffer solution as a background electrolyte, in which indicator, auxiliary and silver chloride reference electrode is placed, removing oxygen in background electrolyte solution by bubbling it with gaseous nitrogen, electron accumulation is carried out, a voltamperogram of the background electrolyte is recorded, then successively adding aliquots of blood serum without prednisone, blood serum with prednisone, standard additives of prednisone solution, removing oxygen after adding each additive, preliminary accumulation of electrons and recording of voltamperograms of electroreduction of prednisone relative to silver chloride reference electrode, determination of prednisone concentration by height of cathode peak by standard additives method, where indicator electrode used is mechanically cleaned disc impregnated graphite electrode; as an auxiliary electrode, a silver chloride electrode is used; a Britton-Robinson buffer solution with pH 7.4 is used, removing oxygen from the background electrolyte solution by bubbling it with nitrogen for 5 minutes, preliminary electron accumulation is carried out for 10 s at potential of +0.8 V, stirring for 10 s and calming solution for 20 s, recording the cathode voltammogram of the background electrolyte solution in the potential sweep range from -1.0 to -1.6 V at rate of 70 mV/s, after adding aliquots of blood serum without prednisone, blood serum with prednisone, standard additives of prednisone solution, removal of oxygen and electron accumulation are carried out under the same above conditions, and recording voltammograms of electroreduction of prednisone is carried out at potential of -1.35±0.05 V in mode of direct-current linear potential sweep.

EFFECT: invention provides developing a voltammetric method for quantitative determination of prednisone in blood serum by cathode voltammetry.
