METHOD FOR DETECTING ANTIBODIES IN BLOOD SERUM ASSOCIATED WITH AUTOIMMUNE ENDOCRINE AND COMORBID PATHOLOGIES ON HYDROGEL BIOCHIP Russian patent published in 2024 - IPC G01N33/53

Abstract RU 2816512 C1

FIELD: medicine.

SUBSTANCE: invention relates to molecular biology, immunology, endocrinology and can be used in clinical laboratory diagnostics for detecting class G antibodies in blood serum, characteristic of autoimmune endocrinopathies, such as autoimmune forms of carbohydrate metabolism disorders (type 1 diabetes mellitus, etc.), autoimmune thyroiditis, autoimmune hypergonadotropic hypogonadism, autoimmune adrenal insufficiency, autoimmune hypoparathyroidism, type 1 autoimmune polyglandular syndrome, as well as antibodies characterizing the development of comorbid pathologies, including autoimmune gastritis, vitiligo, alopecia, celiac disease. Method includes providing a hydrogel biochip with immobilized antigens, such as glutamic acid decarboxylase, insulin, proinsulin, insulin receptor, zinc transporter 8, tyrosine phosphatase, pancreatic islet antigen, thyroperoxidase, thyroglobulin, sodium iodine symporter, 21-hydroxylase, follicle-stimulating hormone receptor, 3β-hydroxysteroid dehydrogenase, 17-hydroxylase, a cholesterol side chain cleavage enzyme, protein 5 containing a leucine-rich NACHT repeat, interferon omega, interferon alpha-2-a, interleukin 22, internal Kastl factor, alpha subunit of H+/K+-ATPase of gastric parietal cells, trichogyline, keratin 16, tyrosinase, dopachrome-tautomerase, transglutaminase 2. Method also includes interaction of a sample of human blood serum with elements of biochip with formation of specific binary immune complexes of antigens, immobilized in elements of biochip, with antibodies in a blood serum sample, detection of formed immune complexes, recording and interpretation of analysis results on a biochip.

EFFECT: invention enables higher efficiency of the method for detecting antibodies in blood serum associated with autoimmune endocrine and comorbid pathologies on a hydrogel biochip.
