FIELD: molecular biology; medicine.

SUBSTANCE: invention relates to molecular biology, immunology, endocrinology and can be used in clinical laboratory diagnostics to determine the avidity index of autoantibodies – immunoglobulins of class G in blood serum. Method is based on multiplex immunoassay on hydrogel biochips containing immobilized proteins – autoantigens. Method comprises steps of incubating a blood serum sample on a biochip to form complexes of immobilized autoantigens with autoantibodies, adding a chaotropic agent to a biochip and detecting immobilized autoantigen-autoantibody complexes using fluorescent-labelled anti-species human immunoglobulin G antibodies. Autoantibody avidity index is calculated as a percentage of the fluorescent signal of the biochip element containing the corresponding autoantigen, in an analysis involving a step for treatment with a chaotropic agent, to the biochip element fluorescent signal obtained without such processing.

EFFECT: method enables to determine the autoantibody avidity index in human blood serum on hydrogel biochips; determination of avidity of autoantibodies can be an additional characteristic allowing to increase specificity of immunoassay.
