METHOD FOR COUNTING REACTIVE NEUTROPHILS IN PERIPHERAL BLOOD ON AUTOMATIC ANALYZER WITH FLUORESCENT FLOW CYTOMETRY TECHNOLOGY Russian patent published in 2024 - IPC G01N33/49 G01N35/08 G01N15/00

Abstract RU 2818470 C1

FIELD: medicine; clinical laboratory diagnostics.

SUBSTANCE: invention can be used for counting reactive neutrophils in peripheral blood on an automatic analyzer with fluorescence flow cytometry technology. Peripheral blood samples are measured on an automatic hematology analyzer with technology of measuring leukocytes by fluorescence flow cytometry for measurement in whole blood (WB) mode. During measurement of leukocytes in a peripheral blood sample in WB mode, the following data are obtained: WBC—total number of leukocytes, 109 cells/l; NEUT #—absolute neutrophil count, 109 cells/l; NEUT % - relative number of neutrophils, in %; IG #—absolute number of immature granulocytes, 109 cells/l; IG %—relative amount of immature granulocytes, in %. In the process of calculations, the neutrophil population span is calculated along the Y axis NE-width-Y, the nuclear shift index of the NF, the relative number of reactive neutrophils RE-NEUT %, absolute number of reactive neutrophils RE-NEUT #, relative number of all immature neutrophils TOTAL IG %, absolute number of all immature neutrophils TOTAL IG #, relative number of segmented neutrophils SEG %, absolute number of segmented neutrophils SEG # in peripheral blood. First, the neutrophil population span is determined along the Y axis NE-width-Y in peripheral blood according to claimed formula (1). Further, a peripheral blood neutrophil nuclear shift index (NSI) is calculated by formula (2). At the same time the value of NSI as per results of calculation (2) takes 2 possible versions: if NSI ≤ 0 and if NSI > 0. Further, the relative amount of reactive neutrophils is determined by claimed formula (3). Value of RE-NEUT % based on the results of calculation (3) takes 2 possible versions: if RE-NEUT % ≤ 0 and if RE-NEUT % > 0. Further, RE-NEUT #, TOTAL IG %, TOTAL IG #, SEG % and SEG # are calculated by formulas (4), (5), (6), (7), (8), respectively.

EFFECT: method provides the possibility of increasing the rate and accuracy of counting and evaluating neutrophil left shift and activation of neutrophils in peripheral blood due to fully automated measurement of absolute and relative number of reactive neutrophils, value of nuclear shift index, total number of all immature neutrophils and number of segmented neutrophils in whole blood samples.
