FIELD: biotechnology.

SUBSTANCE: disclosed is use of a DNA fragment comprising a nucleotide sequence SEQ ID NO: 1 or a sequence having at least 80% homology thereto and identical functional activity, as a promoter in Yarrowia lipolytica yeast. Also disclosed is a yeast transformant Yarrowia lipolytica producing β-carotene and containing in the chromosome at least one copy of the nucleotide sequence SEQ ID NO: 1 or a sequence having at least 80 % homology thereto and exhibiting promoter activity, as well as at least one copy of each of the following optimized synthetic genes: CarRP M. circinelloides, nucleotide sequence of which is given in the sequence list under the number SEQ ID NO: 2, coding bifunctional enzyme phytoene synthase/lycopene P-cyclase, GGPPs7 Synechococcus sp., nucleotide sequence of which is given in the sequence list under the number SEQ ID NO: 3, coding geranylgeranyl pyrophosphate synthase, CarB Mucor circinelloides, nucleotide sequence of which is given in the sequence list under the number SEQ ID NO: 4, coding phytoendehydrogenase. Also disclosed is a recombinant yeast strain Yarrowia lipolytica VKPM Y-5032, which is a producer of β-carotene.

EFFECT: invention provides effective preparation of Yarrowia lipolytica transformant producing β-carotene.
