SUBSTANCE: invention relates to biotechnology and genetic engineering and represents a promoter/enhancer system for increasing the binding of RNA polymerase 1 with promoter regions, consisting of a translational elongation factor promoter 1α (alpha EF1) linked to 3'-end of the cytomegalovirus enhancer, and a ubequitin C promoter linked to 5'-end of the cytomegalovirus enhancer. Invention relates to an expression vector containing a target protein gene under the control of said system, where the target protein gene is two copies of the darbepoetin alpha gene.

EFFECT: invention enables to effectively implement a method for producing a stable clone of darbepoetin alpha, involving transfection of a cell line with the disclosed vector, selection of a stable darbepoetin producer by selection in a medium with subsequent cloning, cultivation of a stable clone in a nutrient medium.
