FIELD: biotechnology.
SUBSTANCE: disclosed is recombinant plasmid DNA p3.4-P4A1IgA2m1mut2-J12His-Intht, coding neutralizing monoclonal antibody P4A1 to SARS-CoV-2 virus of mutant IgA2m1 isotype and human J-chain. Plasmid contains an intronated antibody heavy chain gene, under the control of the hEF1-HTLV promoter and the bovine growth hormone polyadenylation signal BGH polyA, the antibody light chain gene and the human J-chain gene, under control of CMV promoter and internal ribosome binding site (IRES) of encephalomyocarditis virus (EMCV), post-transcriptional regulatory element WPRE of marmot hepatitis virus (WHV) and polyadenylation signal of thymidine kinase of herpes virus. HEF1-HTLV and CMV promoters are arranged in a sequence providing unidirectional transcription. Also disclosed is a method of producing a dimer of a neutralizing monoclonal antibody P4A1 against the IgA2m1 mutant isotype virus with a J-chain, involving culturing CHO cells transfected with recombinant plasmid DNA p3.4-P4A1IgA2m1mut2-J12His-Intht.
EFFECT: inventions provide simultaneous expression of genes of heavy and light chains of antibody and gene of J-chain as a result of transfection with one plasmid and output of neutralizing monoclonal antibody dimer of 4_7 mg/ml of culture medium.
