FIELD: biotechnology.
SUBSTANCE: what is presented is a recombinant plasmid DNA p3.4-P4A1IgA1-J12His-ht coding a neutralizing monoclonal antibody P4A1 to the SARS-CoV-2 virus of the IgA1 isotype and a human J-chain. Plasmid DNA contains a spliced antibody heavy chain gene, under the control of the hEF1-HTLV promoter and the bovine growth hormone polyadenylation signal BGH poly, antibody light chain gene and human J-chain gene under control of CMV promoter and internal ribosome binding site (IRES) of encephalomyocarditis virus (EMCV), post-transcriptional regulatory element WPRE of marmot hepatitis virus (WHV) and polyadenylation signal of thymidine kinase of herpes virus. Promoters of hEF1-HTLV and CMV are arranged in a sequence providing unidirectional transcription. Also described is a method of producing a dimer of neutralizing monoclonal antibody P4A1 against SARS-CoV-2 virus (2019-nCoV) of the IgA1 isotype with J-chain, involving culturing CHO cells transfected with plasmid DNA p3.4-P4A1IgA1-J12His-ht.
EFFECT: inventions provide simultaneous expression of genes of heavy and light chains of antibody and gene of J-chain as a result of transfection with one plasmid and output of neutralizing monoclonal antibody dimer 5_2 mg/ml of culture medium.
