SUBSTANCE: invention relates to pharmaceutical and food industry and can be used to produce a mixture of ethyl esters of iodinated and non-iodinated fatty acids of pharmaceutical quality, having a wide range of applications as diagnostic and therapeutic agents, as well as in the form of food additives. Disclosed is a method for synthesis of a mixture of ethyl esters of iodinated and non-iodinated fatty acids from vegetable oil, which consists in carrying out several stages of processing vegetable oil obtained by cold pressing or by supercritical CO2 extraction. At first step, iodination-hydrolysis of triglycerides of unsaturated fatty acids is carried out in one-pot mode by emulsifying vegetable oil in phosphoric acid, removal from reaction mixture and reactor space of oxygen, loading at temperature of 0°C alkali metal iodide and subsequent temperature increase to 60–100°C and stirring mixture for 20–24 hours. Thereafter, the obtained mixture is purified from residual iodine by separating the organic layer from water and treating it with an aqueous solution of sodium thiosulphate. Further, non-acid ballast substances are removed by neutralizing the organic layer with an aqueous solution of an alkali metal hydroxide to obtain a mixture of alkali metal salts of iodinated and non-iodinated fatty acids, followed by extraction of the ballast substances with an organic solvent. Then salts of iodinated and non-iodinated fatty acids are returned to acid form by treatment with solution of mineral acid and then reaction of esterification of mixture of purified salts of iodinated and non-iodinated fatty acids is carried out under action of ethyl alcohol in presence of mineral acid. After that, final purification of the obtained mixture of ethyl esters of iodinated and non-iodinated fatty acids from impurities of resinous and other undesirable substances is carried out by flash chromatography on silica gel using a mixture of organic solvents as an eluent, subsequently removed by vacuum distillation at a temperature of not higher than 40°C.
EFFECT: obtaining a mixture of esters of iodinated and non-iodinated fatty acids of pharmaceutical quality, having high purity and corresponding to key quality indicators, namely: appearance, weight content of iodine, radiopacity, viscosity, with simultaneous simplification of technology and reduction of cost of synthesis of these esters.
