FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, specifically to polypeptides based on a variant of the Fc region of dog IgG, and can be used in veterinary science in therapy of pain and atopic dermatitis in dogs. Disclosed are polypeptides containing versions of the Fc region of a dog IgG with replacement by Arg or His in a position which corresponds to the amino acid position 434 of the Fc region of a wild type IgG, or with substitutions with Met at a position which corresponds to the position of amino acid 252 of the Fc region of the wild-type IgG, and by His at the position which corresponds to the position of amino acid 434 of the Fc region of the wild-type IgG, wherein the amino acid positions are based on the EC numbering. Wild-type dog IgG is selected from (a) dog IgG.A containing an Fc domain with SEQ ID NO: 9; (b) a dog IgG.B containing an Fc domain with SEQ ID NO: 10; (c) a dog IgG.C comprising an Fc domain with SEQ ID NO: 11; and (d) dog IgG. containing an Fc domain with SEQ ID NO: 12.
EFFECT: invention enables to obtain a polypeptide with higher binding affinity to dog FcRn compared to a control polypeptide containing a wild-type dog IgG Fc region.
