FIELD: chemistry.
SUBSTANCE: invention relates to methods of producing N-monofluoroalkyltropane, trialkyltin tropane, iodinated tropane, as well as to use of N-monofluoroalkyltropane as a precursor for producing trialkyltin tropane and/or iodinated tropane. Method of producing N-monofluoroalkyltropane of formula (VI) involves steps (a) - (f). At step (a), a compound of formula (I) is provided, after which at step (b) its carbonyl group is reduced to a hydroxyl group to obtain a compound of formula (II). At step (c), dehydration of compound (II) is carried out using 4-dimethylaminopyridine, triethylamine and trifluoroacetic acid anhydride to obtain a compound of formula (III). At step (d), 4-bromophenyl fragment is attached to compound (III) to obtain compound (IV). At step (e), N-demethylation of compound (IV) is carried out to obtain a mixture containing the compound of formula (V) and its enantiomer. At step (f), in the presence of a base, the derivative (V) is alkylated with F-(CH2)n-Br, where n is 2, 3 or 4, to obtain a mixture containing N-monofluoroalkyltropane of formula (VI) and its enantiomer , wherein the enantiomers are separated once after step (f).
EFFECT: providing a method of producing N-monofluoroalkyltropanes with a reduced number of synthesis steps, excluding the use of cocaine as a starting material.
