SQUALENGOPENCYCLASE (SHC) (VARIANTS) Russian patent published in 2024 - IPC C12N9/90 C12P17/04

Abstract RU 2829857 C1

FIELD: biotechnology.

SUBSTANCE: invention relates to biotechnology and is a method for producing (-)-ambrox in isomerically pure form or in a mixture, including the enzymatic conversion of (3E, 7E)-homopharnesol (EEN) or a mixture of homopharnesol isomers containing EEN into (-)-ambrox in isomerically pure form or in a mixture, using a variant of the enzyme SHC (squalene-hopene cyclase)/NAS(homopharnesol-ambrox cyclase), where the variant of the enzyme SHC/HAC has an amino acid sequence having at least approximately 90.0% identity with SEQ ID NO: 1, where the amino acid sequence of the variant of the enzyme SHC/HAC has amino acid modifications relative to SEQ ID NO: 1 in the positions corresponding to positions 132, 224 and 432 in SEQ ID NO: 1, which are M132R, A224V and I432T, respectively, and where the amino acid the sequence of the SHC/HAC enzyme variant has amino acid modifications relative to SEQ ID NO:1 at a position corresponding to position 557 in SEQ ID NO: 1, and at least one position corresponding to position 81, 431 or 613 in SEQ ID NO:1, where one or more amino acid modifications at positions 81, 431, 557 or 613 are substitutions, preferably non-conservative substitutions.

EFFECT: invention makes it possible to effectively obtain (-)-ambrox.
